Hans Molisch was a Austrian botanist that create the Molisch's test, that is used to indicate the presence of carbohydrates by the dehydratation of the crabohydrate by sulfuric acid producing an aldehyde molecule that creates a violet ring in the test tube. The hypothesis is the study of carbohydrate concentration in leaves and how it can be test with the analyze of the reaction between the carbohydrate and an acid to form a aldehyde in a ethanol 95% solution.
To make the test the leaves are in the dark to stop the photoshynthesis process and ensure a more accurate value of carbohydrate concentration in rest stage (without the production of new sugars from metabolic source).
2. The leaf is boiled in alcohol solution to dehydrate the tissue and make the reaction happen faster between the carbohydrates and the acid molecule, 3. being used iodine was a dye for the clearest visualization of the experiment.
