DNA ligase can still recircularize the linearized plasmid vector even without an insert DNA fragment, but the resulting plasmid will not have an insert and will be identical to the original uncut plasmid. However, the efficiency of the ligation reaction may be lower in the absence of an insert DNA fragment.
