Jamestown and Plymouth were both permanent English colonies. They both didn't start out as royal colonies. Both were started in the early 1600s (Jamestown=1607, Plymouth=1620) Colonists at Jamestown and Plymouth suffered greatly in the beginning- the colonists at Jamestown weren't used to hard labor they had to do and suffered from famine/illness/ect while the colonists at Plymouth arrived later during both the same year and suffered from an extremely harsh winter. Also, a conflict between them included economical issues. Plymouth's issues included economy and religion, and while Plymouth wasn't dependent, Jamestown was.
Hope this helps (:

Jamestown's population consisted of Europeans and in Plymouth, they had pilgrims. Conflict in Jamestown was mostly concerning its economical issues. Plymouths main issues were economy and religion. Jamestown was dependent on the indians , where Plymouth was not .
