The kind of poison that would most likely directly interfere with glycolysis is a metabolic substance which closely resemble glucose in structure but which can not be metabolized in the same way as glucose. This type of substance will compete will glucose for the active site of hexokinase, which is the first enzyme in the glycolysis process and prevent glucose from binding to the enzyme thus, preventing glycosis process.
