Why do microbiologists need to isolate bacterial colonies from a specimen?

The specimen are composed of numerous bacterial colonies. I order to identify the bacteria, it has to be isolated from the specimen so that it may be cultured in the artificial media and the microbial colony forming units may be counted and identified, for identification of the disease.
