Macrophages are white blood cells that roam the body searching for invading microbes. inside macrophage vacuoles these invaders are destroyed. how do the microbes get inside the macrophages?

endocytosis/phagocytosis. the plasma membrane forms a vesicle around the microbe, dragging (???) it into the cell. the vesicle either forms into a vacuole or transports the microbe to a vacuole where it's destroyed
