The prime activity of DNA polymerase is to synthesize DNA from nucleotides, that is, the building blocks of DNA. There are many kinds of DNA polymerase, which does an essential role in the replication of DNA and they generally function in pairs to duplicate one molecule of double-stranded DNA into two novel double-stranded molecules of DNA.

Apart from developing new copies of DNA, the DNA polymerase is accountable for proofreading the strand so that the misplaced or the wrong base pairs can be corrected. This maintains the veracity of the original strand of DNA, which is passed onto the daughter cells.
