These types of drugs work in the brain, more precisely in the central nervous system (CNS) through the control of neurotransmitters ("messenger-like" chemicals of the brain).

Depressants act by decreasing brain activity, while stimulants accelerate brain activity, both by regulating the activity of neurotransmitters(increasing or decreasing).
