Can a fraction have a negative numerator and a positive denominator

Yes it is possible.

For example: We have have the fraction
(-3)/(4) where the 3 up top is negative and the 4 down below is positive. The overall fraction will simplify to negative three-fourths. When we divide negative over positive, the result is negative.
