For anyone taking the test I believe the answer is:

He declared that it was unconstitutional for a state to label citizens based upon their color with the intent of restricting their privileges.

Homer Plessy's defense was based on that Jim Crow laws violated the Equal Protection Clause of the Fourteenth Amendment and the Thirteenth Amendment which bans slavery. The Equal Protection Clause of the Fourteenth Amendment states that it is forbidden to deny someone within their jurisdiction equal protection of the law, essentially stating that the Jim Crow laws were in violation of the constitution.
