In a subject-by-subject comparison essay, a thesis statement must clarify the main idea or main point and create a focused argument.

A subject-by-subject comparison is basically two separate essays linked with transitions; you state everything you have to say about one subject before transitioning to the next subject. The thesis of a subject-by-subject comparison should clearly state the main idea and/or argument of the essay.
