The ER signal sequence refers to the amino acid sequence in the protein that gets determined by the SRP or the signal recognition particle. This then gets bind with the SRP receptor in the endoplasmic reticulum to help in the insertion of protein to the membrane of the endoplasmic reticulum.

In case, when the ER signal sequence is absent then the complex of the ribosome and the mRNA and the protein getting translated stays in the cytosol. This will eventually result in the release or discharging of protein into the cytosol.
