As the Order of Operations tells you, start with the inner brackets and work outward. Per the distributive property, a minus sign in front of brackets changes the sign of every term inside.

2x -{y +3z -[x +3 -(2 +y) -4] +[-3z -5] +4z}

= 2x -{y +3z -[x +3 -2 -y -4] -3z -5 +4z}

= 2x -{y +3z -[x -y -3] -3z -5 +4z}

= 2x -{y +3z -x +y +3 -3z -5 +4z}

= 2x -{-x +2y +4z -2}
