x - 4y = -4

-4y = -x - 4

y = x/4 + 1

so the slope is 1/4.

the other line is

y = 4x - 4

the form is right, the slope is 4 or 4/1.

the slopes are different, so the lines are not parallel.

a perpendicular slope turns the original slope upside-down AND flips the sign.

a perpendicular slope to the first 1/4 slope would be

-4/1 = -4

but that is different from the second slope.
