Let's suppose that a person is going up a ramp. Due to the change in height, this means that potential energy is increasing and since there is a velocity involved this means that there is also kinetic energy being used.
Another example is an object falling. In this case, when the object is at the highest point its potential energy is maximum. When it starts falling the potential energy is transformed into kinetic energy as the velocity of the object increases. When the object reaches the ground its potential energy is a minimum and the kinetic energy is a maximum but the sum of the kinetic and potential energies remains the same throughout all the process.
