Yes it is. "Mi cuarto es más grande que el cuarto de mi hermana" or "Mi cuarto es más grande que el de mi hermana."

Are you trying to say? "My room is bigger than my sisters room." If so then yes, "Mi cuarto es mas grande que el cuarto de mi hermana." is correct.
